There are lot of factors that prevent the proper functioning of the enzyme.

1.High temperature can cause the denaturing of the enzyme.
2.Slight change in the pH can cause the ionization of the amino acids at the active site.
3.Inhibitor can also retards the rate of reaction.

What make enzymes function properly

Temperature. Enzymes work at a specific temperatures. Enzymes also work at a specific pH. So if the enzyme doesn't get those pH and temperature it will not function properly.

A temperature that is too high- this is because enzymes are proteins, they contain hydrogen bonds, when the temperature is too high these bonds are broken and this causes the tertiary structure to change and thus the active site.

The enzyme has become denatured and it can no longer bind to the substrate with the complementary shape

Secondly the wrong PH. This can mess with the hydrogen bonds because alkali's contain OH- ions and acids contains H+ ions, again this will alter the tertiary structure of the enzymes, change its active site so that the substrate with the complementary shape is no longer able to fit .
